    报告人简介: Jasmonic acid and methyl jasmonate, collectively called jasmonates (JAs), are important signal molecules that regulate gene expression in plant development and defense responses to herbivore attack and mechanical wounding. Genetic analysis of JAs biosynthesis mutants gave evidences that the allene oxide synthase (AOS) is a major control point in the biosynthetic pathway of JAs. Specific inhibitors of AOS are, therefore, useful for dissecting plant defense signaling pathways, as well as regulation mechanism of plant growth and development. In this presentation, I will describe the development of specific inhibitors targeting on the AOS, a key enzyme that catalyzes the first step in the specific pathway of JAs biosynthesis. The AOS inhibitor was designed on the basis of catalytic cycle of AOS. The inhibitory activity of synthesized compounds was evaluated by using recombinant AOS from Arabidopsis expressed in E. coli. Structure-activity relationship studies revealed a potent AOS inhibitor, JM-8686, with an IC50 approximately 10 nM. Steady-state kinetic analysis on the inhibition mechanism of JM-8686 indicated that it was a competitive inhibitor of AOS. In vivo application studies of this synthetic series are now in progress.
